Testosterone Propionate is a synthetic androgen drug. Its structural characteristics are white crystals or off-white crystalline powder, and it is odorless. It is readily soluble in organic solvents such as ethanol and ether, but insoluble in water. Testosterone propionate is an esterified derivative of testosterone, a propionate ester containing 17β -hydroxyandroster-4-ene-3-one.
In terms of synthesis, testosterone propionate can be produced by the reaction of testosterone with propionic acid under acidic conditions. After entering the body, it is first converted into testosterone and then inactivated in the liver. Its metabolic products include androsterone, isoandrosterone and procholanolone, which are mainly excreted from the body in the form of conjugated substances through urine.

 

Testosterone propionate, as an androgen drug, has distinct characteristics of action. It can effectively promote the development and maturation of male organs and secondary sexual characteristics, enhance the anabolism of muscles and bones, and inhibit the effect of estrogen. Its intramuscular injection effect lasts for a long time, usually once every 2 to 3 days is sufficient. In addition to being used to treat androgen deficiency, testosterone propionate can also be used in the treatment of functional uterine bleeding and breast cancer and other conditions. However, excessive use may lead to adverse reactions such as masculinization in women, edema, liver damage, jaundice and dizziness. Therefore, this drug should be contraindicated in pregnant and lactating women as well as patients with prostate cancer.

 

From the perspective of the research and development background, the research and development of testosterone propionate began in the 1930s. At that time, scientists were dedicated to finding ways to prolong the duration of testosterone’s action in the body, in order to enhance its therapeutic effect and facilitate patients’ use. After years of research, German chemists Ernst Laqueur and Hans Hobermann successfully synthesized testosterone propionate for the first time in 1935 and confirmed its androgenic activity in animal experiments. Subsequently, in 1937, Schelling AG of Germany launched the first testosterone propionate product, Testoviron, which became the first commercially available injectable testosterone ester in the US market. Although the use of testosterone propionate has decreased with the development of other testosterone esters such as testosterone heptanoate and testosterone cyclovalpropionate, it still retains its market share in some countries and regions
